  • It was found that an altered dihydrofolate can not be adduced wild type thy+ and thy- extracts were examined. It was found that that the Km values for both their dihydrofolate-reductase activity was nearly the same (figure 1) as where the effects of KCl, CaCl2 and other organic mercurials (Table 1) as was the overall activity levels of dihydrofolaate-reductase. Serine hydroxymethylase and N5N10-methylene dehydroxgenase activities were also similar between the wild type and the mutant.
  • The correlation between the loss of thymidylate synthetase and resistance to inhibitors of dihydrofolate reductase may be due to the one striking difference between thymidylate synthetase.
  • thy- mutants 15T- and B3 were found to be resistant to trimethoprim though 15T- was found to more resistant, the main difference between them being that 15T- has no thymidylate synthetase activity while B3 had at least 20 percent of the normal activity.
